スーパーエンハンサー

Sundey meetingで、深澤先生が「An oncogenic super-enhancer formed through somatic mutation of a noncoding intergenic element. Science. 2014 Dec 12;346(6215):1373-7. 」を抄読してくれました。恥ずかしながら、super-enhancerという言葉自体を知りませんでしたが、「Cancer. Cancer by super-enhancer. Science. 2014 Dec 12;346(6215):1291-2.」もわかりやすく解説してくれました。T細胞の急性リンパ性白血病において、small insertionによりnon-coding RNAのmutationが起こります。これがTAL1 oncogeneの発現を増強する転写因子MYB と他の分子との結合を促進します。super-enhancerというのは、ChIP-seq法によって証明された、近接したゲノム上でこのような多数の調節因子が結合しているenhancerのことでした（What are super-enhancers? Nat Genet. 2015;47:8-12.）。今後、肺癌分野でも発癌に影響を及ぼすnon-coding RNAのmutationがでてくるのでしょうね。
